Characteristic Alteration and Use of Tissue Culture Technique in Carrot Breeding

Abstract
This study was conducted to know the degree of inbreeding depression by self pollination to get high purity of breeding lines and the feasibility of plant tissue culture technology for the purpose of carrot breeding. By self pollination, breeding lines showed severe inbreeding depression on such characteristics as leaf and root weight. In certain breeding line, leaf or root weight decreased to 40% and 41% of original lines, after 2 generations of self pollination. In case of leaf blade segment culture, there were no variations from the regenerated plantlets. And it was possible to select plants with good characteristics after harvesting of transplanted roots, even though root shape was abnormal by the transplanting of tissue cultured plants. In field, this method was supposed to be utilized for stable maintenance of characteristics in true breeding varieties and for maintenance and multiplication of promising plants selected by performance test of breeding lines during spring season and for hybrid seed production, resulting in less inbreeding depression of male parents by this technique.
